Mechanism
Delta Sleep Inducing Peptide (DSIP) is a natural nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first isolated from rabbit cerebral venous blood in 1974. It influences sleep architecture primarily by modulating delta-wave (slow-wave, stage 3–4) sleep, acting on hypothalamic and limbic structures via receptors that remain only partially characterised. DSIP also exerts regulatory effects on the HPA axis — attenuating stress-driven cortisol and ACTH hypersecretion — and influences circadian rhythms of locomotor activity, intracerebral neurotransmitters, and plasma protein concentrations. Its amphiphilic structure enables ready passage through the blood-brain barrier.

DSIP administered IV over 10 days (5 daily + 5 every 48–72h) to 7 patients with chronic migraine, tinnitus, and psychogenic pain significantly lowered pain levels in 6/7 patients and produced a simultaneous significant reduction in concomitant depressive states.

Protocol
How to use it.
Dosing
100–500 mcg subcutaneous injection, administered 30–60 minutes before bed. No standardised guidelines exist. Some practitioners use DSIP only on nights of acute sleep disruption rather than nightly; others use short cycles.
Cycle
2–4 week cycles are typical, with breaks of equal duration. Can also be used as-needed for travel, schedule disruption, or acute insomnia rather than as a nightly protocol.
Contraindications
When to skip it.
Limited long-term human safety data. Avoid in pregnancy. Use caution in individuals with HPA axis disorders (Cushing's, Addison's) or those on corticosteroid therapy. Research on DSIP has been conflicting — Professor Borbély called results 'negative or ambiguous'; the original researcher (Schoenenberger) attributed negative findings to funding gaps rather than inefficacy.
