CJC-1295 No DAC (Modified GRF 1-29) is a 29-amino acid synthetic analogue of endogenous growth hormone-releasing hormone (GHRH) with four strategic amino acid substitutions — including D-Alanine at position 2 for DPP-IV resistance — extending its plasma half-life from ~2 minutes to ~30 minutes. It binds to GHRH receptors (GHRHR) on somatotroph cells in the anterior pituitary, activating adenylate cyclase and increasing intracellular cAMP, which triggers a sharp pulse of GH synthesis and secretion. Without the Drug Affinity Complex (DAC), clearance is rapid, producing discrete pulsatile GH spikes that closely mirror the body's natural circadian GH rhythm rather than sustaining a continuous elevation.

Once-daily administration of CJC-1295 normalised growth and body composition in GHRH knockout mice — demonstrating that even short-acting GHRH analogue pulses can rescue GH-axis deficiency when dosed consistently. Efficacy decreased substantially when dosing intervals stretched to 48–72 hours.

How to use it.
Dosing
100–200 mcg per injection, subcutaneous, administered 1–3 times daily. Most protocols include a pre-bed dose to amplify the body's largest natural nocturnal GH pulse; a pre-workout dose is also common. Frequently combined with Ipamorelin at the same injection site for synergistic GH release.
Cycle
Typical cycles run 8–12 weeks followed by a 4-week break to prevent receptor desensitisation. Results in sleep, energy, and recovery typically emerge within 3–4 weeks; meaningful body composition shifts are observed after 2–3 months of consistent use alongside resistance training and adequate nutrition.
Contraindications

Contraindicated in active malignancy or strong family history of cancer (elevated IGF-1 may act as a growth signal). Caution in diabetics — GH can influence blood glucose.
