Mechanism
AC-Selank-NH2 is an N-acetylated, C-terminal amidated version of Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro), derived from the immunopeptide tuftsin, with modifications that dramatically extend its metabolic half-life versus the parent compound's ~2–3 minute plasma half-life. It acts via two primary mechanisms: (1) positive allosteric modulation of GABA-A receptors — altering the expression of GABA receptor subunits (Gabrb3, Gabre, Gabrq) and GABA transporter Slc6a13 in the frontal cortex to enhance inhibitory tone without sedation; and (2) enkephalinase inhibition — blocking the enzymes that degrade endogenous enkephalins in plasma, thereby elevating natural opioid tone and emotional resilience. It also upregulates BDNF, normalizes monoamine (serotonin, dopamine, norepinephrine) balance, and exerts immunomodulatory effects via IL-6 regulation and Th1/Th2 cytokine normalisation.

What we don't know
Russian clinical trials in generalised anxiety disorder demonstrated efficacy comparable to benzodiazepines (reductions in Hamilton Anxiety Rating Scale scores) with superior tolerability — no sedation, no withdrawal, no cognitive impairment. The N-acetylation dramatically extends metabolic half-life vs parent Selank's ~2-3 min. However, no formal PK study with compartment modelling has been published even in Russian journals. Bioavailability and Tmax for intranasal route are inferred from pharmacodynamic onset (anxiolytic effect within 15-30 min). No Western clinical trials exist.
Why this dosing protocol
Standard dosing mirrors AC-Semax: 200-500mcg intranasal per session, 1-3× daily for sustained anxiolytic effect. SC dosing at 100-300mcg may produce longer duration but slower onset than intranasal. The intranasal route is preferred for rapid CNS access. Split dosing throughout the day is standard for generalised anxiety — morning + afternoon doses maintain consistent anxiolytic coverage without sedation accumulating across doses. Unlike benzodiazepines, there is no tolerance or withdrawal — protocols of 4-12 weeks are common, and long-term use has not been associated with dependence in published clinical data. AC-Selank's enhanced stability allows lower effective doses than standard Selank.

How to use it.
Dosing
200–500 mcg intranasal per session (split dosing, 2–3x daily maintains more stable anxiolytic effect than single dose); or 100–300 mcg subcutaneous injection. AC-Selank-NH2's enhanced stability allows lower effective doses than standard Selank.
Cycle

Contraindications
When to skip it.
