Mechanism
AC-Selank-NH2 is an N-acetylated, C-terminal amidated version of Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro), derived from the immunopeptide tuftsin, with modifications that dramatically extend its metabolic half-life versus the parent compound's ~2–3 minute plasma half-life. It acts via two primary mechanisms: (1) positive allosteric modulation of GABA-A receptors — altering the expression of GABA receptor subunits (Gabrb3, Gabre, Gabrq) and GABA transporter Slc6a13 in the frontal cortex to enhance inhibitory tone without sedation; and (2) enkephalinase inhibition — blocking the enzymes that degrade endogenous enkephalins in plasma, thereby elevating natural opioid tone and emotional resilience. It also upregulates BDNF, normalises monoamine (serotonin, dopamine, norepinephrine) balance, and exerts immunomodulatory effects via IL-6 regulation and Th1/Th2 cytokine normalisation.

How to use it.
Dosing
200–500 mcg intranasal per session (split dosing, 2–3x daily maintains more stable anxiolytic effect than single dose); or 100–300 mcg subcutaneous injection. AC-Selank-NH2's enhanced stability allows lower effective doses than standard Selank.
Cycle
10–14 days most common; up to 30 days in clinical contexts. Follow with 7–14 day washout. No pharmacological dependence develops, but psychological reliance is possible — use as regulatory support, not a crutch.
Contraindications
When to skip it.
Avoid in pregnancy/breastfeeding. Use caution when combining with strong GABAergic medications (benzodiazepines, barbiturates, alcohol) — additive CNS depression possible.
